Maize Transgenes Containing Zein Promoters are Regulated by opaque2

چکیده
Understanding epistatic interactions between transgenes and native genes is critical for predicting the response of transgenes to different genetic backgrounds and environments. Our objective was to determine if predicted epistatic interactions occur between maize (Zea mays L.) transgenes carrying zein promoters and opaque2, a transcriptional regulator of zein genes. Expression of the transgenes was signifi cantly decreased in kernels containing the opaque2 mutation. Native zein proteins in these kernels were reduced as well. Thus, transgene expression mirrored expression of the native gene that contributed the promoter of the transgene. This work demonstrates that information about native gene expression may be useful for predicting transgene expression, reducing the risk of unintended consequences of transgene expression. M.N. Divergent Transactivation of Maize Storage Protein Zein Genes by the Transcription Factors Opaque2 and OHPs
Maize transcription factors (TFs) opaque2 (O2) and the O2 heterodimerizing proteins (OHP1 and OHP2) originated from an ancient segmental duplication. The 22-kDa (z1C) and 19-kDa (z1A, z1B, and z1D) α-zeins are the most abundant storage proteins in maize endosperm. O2 is known to regulate α-zein gene expression, but its target motifs in the 19-kDa α-zein gene promoters have not been identified. ...
متن کاملThe transcriptional activator Opaque2 recognizes two different target sequences in the 22-kD-like alpha-prolamin genes.
The maize Opaque2 (O2) protein is a "leucine zipper" DNA binding factor that interacts with the sequence TCCACGTAGA in the promoters of the 22-kD alpha-zein genes and activates its transcription. A completely different consensus sequence (GATGAPyPuTGPu) identified in b-32, a gene that encodes an abundant albumin that is also under control of the O2 locus, can also be bound by the O2 protein. We...
متن کاملZmMADS47 Regulates Zein Gene Transcription through Interaction with Opaque2.
Zeins, the predominent storage proteins in maize endosperm, are encoded by multiple genes and gene families. However, only a few transcriptional factors for zein gene regulation have been functionally characterized. In this study, a MADS-box protein, namely ZmMADS47, was identified as an Opaque2 (O2) interacting protein via yeast two-hybrid screening. The N-terminal portion of ZmMADS47 contains...
متن کاملWild-type opaque2 and defective opaque2 polypeptides form complexes in maize endosperm cells and bind the opaque2-zein target site.
The Opaque2 (O2) basic leucine (Leu)-zipper transcriptional activator controls the expression of several genes in maize (Zea mays). We investigated the phosphorylation extent of wild-type O2 and mutant-defective or mutant-truncated o2 polypeptides in endosperm cells, their subcellular localization, participation in complex formation, and involvement in functional activity. Besides wild type, fo...
متن کاملIntrogression of opaque2 into Waxy Maize Causes Extensive Biochemical and Proteomic Changes in Endosperm
Waxy maize is prevalently grown in China and other countries due to the excellent characters and economic value. However, its low content of lysine can't meet the nutritional requirements of humans and livestock. In the present study, we introgressed the opaque2 (o2) allele into waxy maize line Zhao OP-6/O2O2 by using marker-assisted selection (MAS) technique and successfully improved the lysin...
